The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
Storage conditions for semaglutide depend heavily on the presentation. Lyophilized research powder is generally kept at two to eight degrees Celsius in a sealed container, protected from light and moisture. Manufacturer labeling for finished injectable products specifies refrigeration before first use, with defined in-use periods at room temperature afterward. The oral tablet form is stored at controlled room temperature and is more tolerant of short excursions. Temperature excursions should be documented rather than inferred.

Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
In general, polystyrene is not accepted in curbside collection recycling programs and is not separated and recycled where it is accepted. In Germany, polystyrene is collected as a consequence of the packaging law (Verpackungsverordnung) that requires manufacturers to take responsibility for recycling or disposing of any packaging material they sell. Most polystyrene products are currently not recycled due to the lack of incentive to invest in the compactors and logistical systems required. Due to the low density of polystyrene foam, it is not economical to collect. However, if the waste material goes through an initial compaction process, the material changes density from typically 30 kg/m3 to 330 kg/m3 and becomes a recyclable commodity of high value for producers of recycled plastic pellets. Expanded polystyrene scrap can be easily added to products such as EPS insulation sheets and other EPS materials for construction applications; many manufacturers cannot obtain sufficient scrap because of collection issues. When it is not used to make more EPS, foam scrap can be turned into products such as clothes hangers, park benches, flower pots, toys, rulers, stapler bodies, seedling containers, picture frames, and architectural molding from recycled PS. As of 2016, around 100 tonnes of EPS are recycled every month in the UK. Recycled EPS is also used in many metal casting operations. Rastra is made from EPS that is combined with cement to be used as an insulating amendment in the making of concrete foundations and walls.
Matrix metalloproteinase-9 (MMP-9), also known as 92 kDa type IV collagenase, 92 kDa gelatinase or gelatinase B (GELB), is a matrixin, a class of enzymes that belong to the zinc-metalloproteinases family involved in the degradation of the extracellular matrix. In humans the MMP9 gene encodes for a signal peptide, a propeptide, a catalytic domain with inserted three repeats of fibronectin type II domain followed by a C-terminal hemopexin-like domain.

== Society and culture == Some commentators argue that skin cancer surgery, including Mohs surgery, is overutilised as rates of skin cancer surgery are increasing worldwide. It is unclear if this relates to higher rates of skin cancer, increased vigilance in diagnosis, and increased availability of the procedure, or patient and doctor preferences. The incidence of Mohs surgery increased significantly over the decade between 2004 and 2014. In a sample of 100 Mohs surgeries, the total cost ranged from US$474 to US$7,594, with the higher costs for hospital-based complex procedures. In Australia, the direct out of pocket cost to patients may vary from $0 to $4000. When the non-Mohs surgery is performed by multiple doctors including pathologists the costs may be increased further. This is especially true when the cancer is incompletely excised and requires repeat surgery.

At 1 atm pressure, it reaches its maximum density of 999.972 kg/m3 (62.4262 lb/cu ft) at 3.98 °C (39.16 °F). Below that temperature, but above the freezing point of 0 °C (32 °F), water expands (becoming less dense) until it reaches the freezing point, at which its density in the liquid phase is 999.8 kg/m3 (62.4155 lb/cu ft). As water cools to 3.98 °C (39.16 °F), water volume decreases. As it freezes and becomes ice, water expands by about 9%, reaching a density of 917 kg/m3 (57.25 lb/cu ft). This expansion can exert enormous pressure, bursting pipes and cracking rocks. As a solid, it displays the usual behavior of contracting and becoming more dense as it cools. These unusual thermal properties have important consequences for life on earth. In a lake or ocean, water at 4 °C (39 °F) sinks to the bottom, and ice forms on the surface, floating on the liquid water. This ice insulates the water below, preventing it from freezing solid. Without this protection, most aquatic organisms residing in lakes would perish during the winter. In addition, this anomalous behavior is an important part of the thermohaline circulation which distributes heat around the planet's oceans.
==== Genetically induced ==== Type 1 diabetes (T1D) is a multifactorial autoimmune disease with a strong genetic component. Although environmental factors also play a significant role, the genetic susceptibility to T1D is well established, with several genes and loci implicated in disease development. The most significant genetic contribution to T1D comes from the human leukocyte antigen (HLA) region on chromosome 6p21. The HLA class II genes, particularly HLA-DR and HLA-DQ, are the strongest genetic determinants of T1D risk. Specific combinations of alleles such as HLA-DR3-DQ2 and HLA-DR4-DQ8 have been associated with a higher risk of developing T1D. Individuals carrying both of these haplotypes (heterozygous DR3/DR4) are at an even greater risk. These HLA variants are thought to influence the immune system's ability to differentiate between self and non-self antigens, leading to the autoimmune destruction of pancreatic beta cells. Conversely, some HLA haplotypes, such as HLA-DR15-DQ6, are associated with protection against T1D, suggesting that variations in these immune-related genes can either predispose or protect against the disease. In addition to HLA, multiple non-HLA genes have been implicated in T1D susceptibility. Genome-wide association studies (GWAS) have identified over 50 loci associated with an increased risk of T1D. Some of the most notable genes include:
=== Peru === Joseph Dombey, in a letter written from Lima on 20 May 1779, specifies the ancestral way used by the Peruvians to prepare potatoes that constitute, along with maize (U.S.: "corn"), their primary foods. Further, that they carry potatoes in haversacks on long journeys, for which the potato is prepared by cooking it in water, then peeling it, and finally exposing it to the wind and sun until it has completely dried. The process can preserve it "several centuries, by guaranteeing it of the humidity". This papa seca (dried potato) is then mixed in with other foods to make meals. Another process consists of freezing the potato and treading on it to remove the skin. Thus prepared, it is put in running water and loaded with stones. Fifteen or twenty days later, it is exposed to the sun until it dries. It becomes the chuño, "a real starch, with which one could make powder for the hair". The Peruvians use it to prepare jams, a flour for convalescents, and mix it with almost all their dishes. An author of the 20th century points out that the process of the Peruvians, who operate by freezing followed by dehydration, is none other than "a freeze-drying by the natural means". He specifies that the tubers are left in frozen water several nights before being exposed to the sun and trodden on and that, "to make the product suitable for consumption, it is enough to put it back in water". According to him, the Spaniards used this preparation in the 16th century to feed the indigenous people forced to work in the silver mines of Potosi.

Battles and skirmishes took place during the Aleutian Islands campaign of World War II. The Japanese landing and occupations of Kiska and Attu, in June 1942, were the only invasions of the United States in North America during WWII; the Philippines, Guam and Wake Atoll, Pacific territories of the United States, were also invaded.
Glycolipid transfer protein is a cytosolic protein that catalyses the transfer of glycolipids between different intracellular membranes. It was discovered by Raymond J. Metz and Norman S. Radin in 1980 and partially purified and characterized in 1982. Recent reviews on structure and possible function are available. This protein transports primarily different glycosphingolipids and glyceroglycolipids between intracellular membranes, but not phospholipids. It might be also involved in translocation of glucosylceramides. It was found in brain, kidney, spleen, lung, cerebellum, liver and heart.
freezing point Also crystallization point. The temperature at which a substance changes state from a liquid to a solid. Because freezing is the reverse of melting, the freezing point of a substance is identical to its melting point, but by convention only the melting point is referred to as a characteristic property of a substance.
== Connection to nitrogen storage in Cyanobacteria == Cyanophycin is highly resistant to degradation by all conventional proteases, and the only enzyme known to be capable of hydrolyzing it is cyanophycinase. Cyanophycin is a non-ribosomally synthesized peptidyl polymer that is used for nitrogen storage by cyanobacteria and other select eubacteria. Approximately 90% of cyanobacteria are diazotrophic, meaning that they can grow without an external source of fixed nitrogen. Diazotrophic growth was severely impaired in bacteria with a mutated cyanophycinase gene, indicating that the inability to degrade cyanophycin is detrimental for the diazotrophic growth of the cyanobacterium, due to an excess of nitrogen storage.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
