Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.

Lyophilised semaglutide is generally held at -20 °C or below, protected from light and moisture. Reconstituted solutions are typically kept at 2-8 °C and used within a defined window because degradation accumulates over time. Repeated freeze-thaw cycles are discouraged, since each cycle can promote aggregation and reduce monomeric content. Room-temperature stability of the solid has been examined in some studies but remains incompletely characterised for long durations, so cold storage is the conservative default for research material.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
RNA-dependent RNA polymerase (RdRp) or RNA replicase is an enzyme that catalyzes the replication of RNA from an RNA template. Specifically, it catalyzes synthesis of the RNA strand complementary to a given RNA template. This is in contrast to typical DNA-dependent RNA polymerases (DdRP), which all organisms use to catalyze the transcription of RNA from a DNA template. RdRp is an essential protein encoded in the genomes of most RNA-containing viruses that lack a DNA stage. Some eukaryotes also contain RdRps, which are involved in RNA interference and differ structurally from viral RdRps. Viral RdRps were discovered in the early 1960s from studies on Picornaviruses when it was observed that these viruses were not sensitive to actinomycin D, a drug that inhibits cellular DNA-directed RNA synthesis. This lack of sensitivity suggested the action of a virus-specific enzyme that could copy RNA from an RNA template.
Common donors in oligosaccharide synthesis are glycosyl halides, glycosyl acetates, thioglycosides, trichloroacetimidates, pentenyl glycosides, and glycals. Of all these donors, glycosyl halides are classic donors, which played a historical role in the development of glycosylation reactions. Thioglycoside and trichloroacetimidate donors are used more than others in contemporary glycosylation methods. When it comes to the trichloroacetimidate method, one of the advantages is that there is no need to introduce heavy metal reagents in the activation process. Moreover, using different bases can selectively lead to different anomeric configurations. (Scheme 2) As to the thioglycosides, the greatest strength is that they can offer temporary protection to the anomeric centre because they can survive after most of the activation processes. Additionally, a variety of activation methods can be employed, such as NIS/ AgOTf, NIS/ TfOH, IDCP (iodine dicollidine perchlorate), iodine, and Ph2SO/ Tf2O. Furthermore, in the preparation of 1, 2-trans glycosidic linkage, using thioglycosides and imidates can promote the rearrangement of the orthoester byproducts, since the reaction mixtures are acidic enough.
Ravenholm is a fictional ghost town in the 2004 first-person shooter game Half-Life 2 created by Valve. It is the setting for the game's sixth chapter, "We Don't Go To Ravenholm...", which follows protagonist Gordon Freeman as he journeys through the area after escaping a Combine attack in order to reach a nearby Resistance outpost. An Eastern European mining town, Ravenholm's residents have turned into hostile zombies due to Combine attacks. The town's sole survivor, Father Grigori, offers his assistance to Freeman throughout the level, culminating in a last stand at the town cemetery. The level received critical praise due to its level design and unexpected usage of horror aspects involving headcrabs and zombies, with some critics calling it one of the most well-designed levels in a Valve game and one of the best first-person shooter levels ever made. Due to its popularity, Valve initially contracted Junction Point Studios to make a prequel featuring the town, a project that later became a spinoff game developed by Arkane Studios that further followed the story of Grigori and his fight against the Combine. However, both projects were ultimately cancelled partway through development.
Dapagliflozin/saxagliptin/metformin, sold under the brand name Qternmet XR among others, is a fixed-dose combination anti-diabetic medication used as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes. It is a combination of dapagliflozin, saxagliptin, and metformin. It is taken by mouth. The drug is marketed by AstraZeneca. The most common side effects include infections of the nose and throat, hypoglycaemia (low blood sugar) when used with a sulphonylurea and effects on the gut such as nausea (feeling sick), vomiting, diarrhoea, abdominal (tummy) pain and loss of appetite. Dapagliflozin/saxagliptin/metformin was approved for medical use in the United States in May 2019, and in the European Union in November 2019. Its marketing authorisation was withdrawn in the European Union in August 2020, and its approval was withdrawn in the US in April 2021, in both cases at the request of AstraZeneca.

Mass resolution can be improved in axial MALDI-TOF mass spectrometer where ion production takes place in vacuum by allowing the initial burst of ions and neutrals produced by the laser pulse to equilibrate and to let the ions travel some distance perpendicularly to the sample plate before the ions can be accelerated into the flight tube. The ion equilibration in plasma plume produced during the desorption/ionization takes place approximately 100 ns or less, after that most of ions irrespectively of their mass start moving from the surface with some average velocity. To compensate for the spread of this average velocity and to improve mass resolution, it was proposed to delay the extraction of ions from the ion source toward the flight tube by a few hundred nanoseconds to a few microseconds with respect to the start of short (typically, a few nanosecond) laser pulse. This technique is referred to as "time-lag focusing" for ionization of atoms or molecules by resonance enhanced multiphoton ionization or by electron impact ionization in a rarefied gas and "delayed extraction" for ions produced generally by laser desorption/ionization of molecules adsorbed on flat surfaces or microcrystals placed on conductive flat surface. Delayed extraction generally refers to the operation mode of vacuum ion sources when the onset of the electric field responsible for acceleration (extraction) of the ions into the flight tube is delayed by some short time (200–500 ns) with respect to the ionization (or desorption/ionization) event.
=== Schizophrenia === In December 2019, the US Food and Drug Administration (FDA) approved lumateperone for the treatment of schizophrenia in adults. Lumateperone is more effective than placebo for reducing positive symptoms after 4–6 weeks of treatment. Some clinical trials report that lumateperone may be effective for treating negative symptoms.
=== Response to injury === Since several immune mechanisms involved in the response to injury are widely shared among modern eukaryotes, it has been suggested that they were present in the LECA. These mechanisms include receptors, calcium signalling, reactive oxygen species, adenosine triphosphate release, kinase cascades, and oxylipin signalling. Single-celled eukaryotes, such as choanoflagellates, substantially share the pathways found in plants and animals for detecting damage and pathogens. Extracellular adenosine triphosphate is a signal that promotes healing of wounds to the epithelium in animals. The signal is detected by a P2X receptor. The cell biologist Thibaut Brunet and the molecular biologist Detlev Arendt propose that the LECA possessed a calcium-based wound healing response. They argue that the mechanism's purpose was to detect and heal a potentially fatal opening in the cell membrane. They propose that it worked by detecting an inflow of calcium ions, which provoked a contraction in muscle-like actomyosin proteins. This in turn caused vesicles to fuse with the cell membrane (exocytosis), healing the opening and preventing the cell from splitting open.
=== 1997–present: CKE, Imasco, Plamondon Companies === In 1997, CKE Restaurants acquired Hardee's from Imasco, but Imasco retained the Roy Rogers trademark and franchise system. The Riese family, which owned 18 Roy Rogers restaurants, sued CKE Restaurants and Imasco for $10 million in 1997, claiming the Roy Rogers chain has been destroyed through "a series of marketing errors of epic proportions." Imasco sold Roy Rogers to Plamondon Enterprises (now Plamondon Companies) in 2002, after three years of negotiation. Roy Rogers was relaunched as Roy Rogers Franchise Company, LLC. Plamondon had already opened the first new Roy Rogers restaurant in Frederick, Maryland, in 2000. At the time of the sale, there were 63 existing Roy Rogers franchises in nine states. In 2021, Roy Rogers announced a strategy to build concentrically out of core markets in the Mid-Atlantic, like Maryland, Virginia, West Virginia, and Pennsylvania. The growth circle would slowly get bigger and include New York, the Eastern Seaboard, and states like Ohio, Tennessee, Florida, Texas, Louisiana, Mississippi, Alabama, and Georgia. The next year, the company announced a new partnership with One Holland Corporation restaurant group and planned to open 10 new Roy Rogers locations over the next six years in the Cincinnati metropolitan area. The new locations would be in Hamilton, Butler, and Clermont counties in Southwest Ohio; Boone, Kenton, and Campbell counties in Northern Kentucky; and Dearborn County in Southeast Indiana. Despite these plans, a spokesperson for One Holland Corp.

The substrates of this enzyme are biopterin, reduced nicotinamide adenine dinucleotide phosphate (NADPH), and two protons. Its products are tetrahydrobiopterin and oxidised NADP+. It was isolated from the parasite Leishmania major. This enzyme belongs to the family of oxidoreductases, specifically those acting on the CH-NH group of donors with NAD+ or NADP+ as acceptor. The systematic name of this enzyme class is 5,6,7,8-tetrahydrobiopterin:NADP+ oxidoreductase. Other names in common use include PTR1, and pteridine reductase 1.
== Depleted uranium == Depleted uranium used in kinetic energy penetrators is supposed to be made from uranium enrichment tailings that have never been irradiated in a nuclear reactor, not reprocessed uranium. It should then contain no detectable amount of uranium-236. However, there have been claims of it being found in some depleted uranium.
===== Sulfonylureas ===== Sulfonylureas act by increasing insulin release from the beta cells in the pancreas. A 2019 systematic review suggested that there is limited evidence if the combined use of metformin with sulfonylurea compared to the combination of metformin plus another glucose-lowering intervention, provides benefit or harm in mortality, severe adverse events, macrovascular and microvascular complications. Combined metformin and sulfonylurea therapy did appear to lead to a higher risk of hypoglycemia. Metformin is available combined with the sulfonylureas glipizide (Metaglip) and glibenclamide (US: glyburide) (Glucovance). Generic formulations of metformin/glipizide and metformin/glibenclamide are available (the latter is more popular).
== Evolution == When a synonymous or silent mutation occurs, the change is often assumed to be neutral, meaning that it does not affect the fitness of the individual carrying the new gene to survive and reproduce. Synonymous changes may not be neutral because certain codons are translated more efficiently (faster and/or more accurately) than others. For example, when a handful of synonymous changes in the fruit fly alcohol dehydrogenase gene were introduced, changing several codons to sub-optimal synonyms, production of the encoded enzyme was reduced and the adult flies showed lower ethanol tolerance. Many organisms, from bacteria through animals, display biased use of certain synonymous codons. Such codon usage bias may arise for different reasons, some selective, and some neutral. In Saccharomyces cerevisiae synonymous codon usage has been shown to influence mRNA folding stability, with mRNA encoding different protein secondary structure preferring different codons. Another reason why synonymous changes are not always neutral is the fact that exon sequences close to exon-intron borders function as RNA splicing signals. When the splicing signal is destroyed by a synonymous mutation, the exon does not appear in the final protein. This results in a truncated protein. One study found that about a quarter of synonymous variations affecting exon 12 of the cystic fibrosis transmembrane conductance regulator gene result in that exon being skipped.

Different techniques detect different classes of impurities, so a single number does not describe a sample completely. Reversed-phase chromatography resolves related peptides well but can miss inorganic salts, while mass spectrometry confirms mass without quantifying everything present. Comparing results requires knowing which method was used and how it was validated.
Cycling between frozen and liquid states can promote aggregation and surface adsorption at the container wall. Each cycle exposes the peptide to transient concentration and pH shifts near the ice interface. Aliquoting before storage limits the number of cycles a single container experiences.
Purity describes how much of the material is the intended substance, while identity describes whether that substance is the correct molecule. Mass spectrometry gives an observed mass that is compared with the theoretical value for the sequence. Peptide mapping after digestion adds sequence-level confirmation that mass alone cannot provide.
Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
